一、个人简介

赵天永,男,教授,博士生导师,无党派人员。2011年到西北农林科技大学生命科学学院生化与分子生物学系工作。

二、研究方向

方向1: 玉米抗旱分子生物学。

植物细胞中的棉子糖(Raffinose)是自然界中最知名的一种三糖,由半乳糖、果糖和葡萄糖结合而成,其在植物中的含量仅次于蔗糖。棉子糖虽然不能被肠道吸收,却是人体肠道中有益菌的营养源和有效的增殖因子,能改善人体的消化功能,促进人体对钙的吸收。从而增强人体免疫力。此外,棉子糖也是种子萌发所需能量的主要来源。对某些植物中棉子糖代谢及功能的研究表明,它可能作为一种渗透调节剂(osmoprotectant)直接参与保护干旱等逆境条件下的细胞功能,因而其含量很可能直接影响植物对逆境的抵抗能力。

玉米中棉子糖相关工作尚未见报道。肌醇半乳糖苷合成酶(Galactinol Synthase, 简称GolS)和棉子糖合成酶(Raffinose Synthase, 简称RafS)是棉子糖合成的两个关键酶。而碱性半乳糖苷酶(a-Alkaline Galactosidase, 简称AGAL)能够水解棉子糖。本项目组已经克隆了玉米肌醇半乳糖苷合成酶和碱性半乳糖苷酶每个基因家族的各三个成员 (ZmGolS1-3 和ZmAGAL1-3)的cDNA并研究了它们在玉米种子萌发和不同发育阶段的表达及对逆境胁迫的响应(Zhao, 2003; Zhao, 2004a; 2004b; Zhao, 2006)。我们最近获得并鉴定了突变子(mutator)插入失活的ZmGolS1-3和ZmAGAL1-3基因的玉米突变体。本实验室的目标是系统研究玉米中控制棉子糖合成的关键酶GolS和RafS基因以及控制棉子糖水解的AGAL酶基因的功能,研究它们在玉米发育过程中以及逆境胁迫条件下在各器官中的表达。同时克隆GolS、RafS基因启动子并鉴定在逆境条件下与其互作的转录因子。最终通过提高在逆境条件下GolS和RafS基因及其转录因子的表达量或降低AGAL基因的表达水平提高玉米的抗逆境能力。这一工作将阐明棉子糖合成途径在玉米耐逆中的作用与机制,并将促进玉米抗逆境分子机理研究和玉米抗逆境育种实践。

方向2:MRP2基因的翻译调控。

MRP2(Multidrug Resistance Protein 2)是一种重要的转运蛋白,对肝脏内某些有机盐和药物的跨膜运输起着非常重要的作用。

我们的前期工作发现MRP2 5'UTR上的uORF(upstream open reading frame) 抑制下游ORF的翻译(Zhang &Zhao, 2010)。而IGF2BP1(insulin growth factor 2 mRNA binding protein 1)作为反式作用因子结合到MRP2 5’UTR 上紧邻起始密码子的一个35核苷酸RNA motif,促进下游ORF的翻译。我们最近发现MRP2的-24C>T SNP (占群体18%)影响相关蛋白因子与35 核苷酸RNA motif 的结合, 抑制下游ORF的翻译。我们将对相关蛋白因子与35 核苷酸RNA motif及其含有-24C>T SNP的RNA motif之间的相互作用,以及这种互作对下游ORF翻译的影响展开研究工作。

三、学习经历

1986-1990: 河北农业大学园艺系果树专业,获学士学位。

1993-1998: 中国农业大学生物化学与分子生物学系,硕博连读, 师从谢友菊教授和王国英教授从事玉米组织培养和遗传转化训练。获作物遗传育种专业博士学位。

2003-2011: 在University of Kentucky工作其间选修了<生物信息学>,<功能基因组>等研究生课程。

四、 工作经历

1990.7–1993.7: 河北科技师范学院园艺系教师,从事果树育种课教学。

1998.10-2000.3: 美国加州大学伯克立分校植物与微生物学系博士后 (Department of Plant and Microbial Biology, U.C.Berkeley),从事玉米基因组研究。

2000.4-2001.9: 美国肯塔基大学园艺系博士后 (Department of Horticulture, University of Kentucky),从事玉米抗逆分子生物学研究。

2001.10-2003.8: 肯塔基大学植物病理系博士后 (Department of Plant Pathology, University of Kentucky),从事真菌病毒学研究。

2003.9-2007.3: 肯塔基大学解剖与神经生物学系Scientist I (Department of anatomy and neurobiology, University of Kentucky),从事爱滋病毒侵染神经细胞的分子机理研究。

2007.4–2011: 肯塔基大学毒理研究中心Scientist II (Graduate Center for Toxicology, University of Kentucky),从事MRP2基因翻译调控研究并兼职于肯塔基大学基因芯片中心(MicroArray Core Facility, University of Kentucky)从事管理和技术支持工作。
